Conditions: Separation of biphosphate adducts: For the development in the D1 direction the PEI-cellulose plates are equipped with a 10x20 cm filter paper which is folded in the middle (5x20 cm) and attached to the PEI-cellulose plates at both sides. They are placed in a chromatography tank (cat. no. 814019) big enough to allow the simultaneous development of a number of PEI-cellulose plates in the D1 direction with 1.0 M sodium phosphate, pH 6.8, overnight. The D2 direction is omitted. The plates are developed in the D3 direction with 4.0 M lithium formate, 6.8 M urea, pH 3.4, in the D4 direction with 0.6 M sodium phosphate, 6.5 M urea, 0.61 M Tris-HCl, pH 8.2, and in the D5 direction with 1.7 M sodium phosphate, pH 6.0. Adduct quantification is based on the spectrophotometric measurement of DNA concentration and the specific activity of [g-32P]ATP.
